3. Introdução
• The great evolutionary pulses of metabolic and structural
innovation of life on Earth can be correlated with changes in
environmental oxygen (HOLLAND, 2006 apud NAVIAUX, 2012).

Estresse Oxidativo
• Decorre da existência de um
desequilíbrio entre compostos
oxidantes e antioxidantes, em
favor da geração excessiva de
radicais livres ou em
detrimento da velocidade de
remoção desses. Tal processo
conduz à oxidação de
biomoléculas com consequente
perda de suas funções
biológicas e/ou desequilíbrio
homeostático, cuja
manifestação é o dano
oxidativo potencial contra
células e tecidos.

Estresse Oxidativo e DCV
20. Estresse Oxidativo e DCV
Figure. Development of atherosclerosis. ROS produced by endothelial cells, SMCs, and macrophages oxidize LDL in the
subendothelial space, at the sites of endothelial damage, initiating events that culminate in the formation of a fibrous
plaque. Rupture of fibrous plaque leads to thrombus formation and occlusion of the vessel. Madamanchi et al, 2005
22. NAD(P)H Oxidase e DCV
• Cell culture studies and animal models
provide evidence for the critical role of
NAD(P)H oxidase-derived oxidative
stress in atherosclerosis ↔ Ang II,
thrombin, PDGF, and TNF-α.
• …the atheroprotective laminar shear
stress→↓NAD(P)H activity
• ...atherogenic oscillatory shear stress→
(+) NAD(P)H activity
• …enhanced AT1 receptor regulation and
increased NADH-dependent vascular O2
-
production were associated with
endothelial dysfunction.
• In human studies... Ang II, AT1,
trombin↔ROS and Ox-LDL↔NAD(P)H
oxidase subunit p22phox: foam cell,
plaque stability and apoptosis in
endothelial and SMCs.
23. Xantina Oxidase e DCV
• XO generates O2
- by catalyzing
hypoxanthine and xanthine to uric
acid.
• XO exists in plasma and endothelial
cells but not in SMCs.
• In the coronary arteries of patients
with coronary artery disease (CAD),
electron spin resonance studies show
significant activation of both NAD(P)H
oxidase and XO
• …endothelial XO is inversely
proportional and positively related to
the effect of vitamin C on
endothelium-dependent vasodilation
• In asymptomatic young individuals
with familial hypercholesterolemia,
the increase of vascular XO activity is
an early event.
24. Metabolismo do AA e DCV
• Lipoxygenases are another important
source of ROS production in the vascular
wall.
• Leukocyte-type 12/15-lipoxygenase (LO)
and its products, 12(S)-
hydroxyeicosatetraenoic acid[12(s)-HETE)]
and 15(S)-HETE, are also implicated in
atherogenesis.
• 12/15-LO activation leads to SMC growth,
hypertrophy, and inflammatory gene
expression; SMCs deficient in the enzyme
show reduced generation of mitogen-
induced ROS.
25. NOS desacoplada e DCV
• Increased vascular O2
- production not only
alters endothelium-dependent vascular
relaxation through interaction with NO, but
the resultant peroxynitrite can also oxidize
BH4.
26. ...during mitochondrial oxidative
phosphorylation under pathophysiologic
conditions, the lectron transport chain may
become uncoupled, leading to increased O2
-
production.

Mitochondrial DNA (MtDNA) is prone to
oxidative damage.

Estresse Oxidativo na Hipertensão
28. • …antioxidants prevent the progression of several pathological
processes – such as cardiac hypertrophy, cardiac myocyte apoptosis,
ischemia-reperfusion, and myocardial stunning…
• TNF-α and Ang II induced hypertrophy in a ROS-dependent
manner…
• Overexpression of catalase significantly reduced Ang II-induced
hypertrophy, and transfection with antisense p22phox inhibited
Ang II–induced H2O2 production.
• During compensated hypertrophy... NAD(P)H oxidase-dependent
ROS production significantly and progressively increased to a peak
at the level of decompensated heart failure.
• ...increased peroxynitrite may cause cytokine-induced myocardial
contractile failure by inactivating sarcoplasmic Ca2+-ATPase and
dysregulating Ca2+ homeostasis.
