Analysis of mRNA levels of ARF6 & ARF8
RT-PCR
Conclusion:- miR167 can indeed reduce the level of ARF8, but not ARF6 transcripts, in
transgenic plants that carried a 35S::MIR167b fusion.

6. • Since group I 35S::MIR167b transgenic plants produced stamens with a
short filament and an undehisced anther . These phenotypes provide
explanations for the reduced fertility in the 35S:: MIR167b transgenic
plants ( Nagpal et al.,2005).

To determine whether pollen grains of the 35S::MIR167b anther were functionally
defective, they performed reciprocal crosses between wild-type and 35S::MIR167b
plants.
They also did pollen germination test in which they found that rate of germination was
much lower in transgenics than that was in wild type
To determine whethe the increasedexpression of miR167 cause degradation of transcripts from ARFs,,, they measured the mRNA levels of the two ARFs by RT-PCR in transgenic lines and wild type plants. Comparison of mRNA levels of wild type and lines reveals degradation of ARF8 transcripts in transgenics although there was no such effect on ARF6 transcrpits.
To understand better the role of miR167 and functions of ARF6 & ARF8, they did phenotypic characterization of transgenic lines and compared it with wild type. Thus this miRNA affects the normal plant development and its physiology.
