Generic Orlistat Capsules are used along with a mildly
hypocaloric diet for the treatment of obese patients with a
body mass index (BMI) greater or equal to 30 kg/m, or
overweight patients (BMI greater or equal to 28 kg/m)
with associated risk factors.
Treatment with Generic Orlistat capsules should be
stopped after 12 weeks if patients have been unable to
lose at least 5 % of the body weight as measured at the
start of therapy.

Chemical Structure - Orlistat

The active ingredient contained in
Generic Orlistat capsules is Orlistat.
Each Generic Orlistat capsule contains
60 mg or 120 mg of Orlistat.

Uses of Generic Orlistat Capsules 60 mg and 120 mg Capsules:
Generic Orlistat is a medication for chronic weight management. It is for
people with overweight and weight-related complications or obesity.
Generic Orlistat capsules are indicated for obesity management including
weight loss and weight maintenance when used in conjunction with a
reduced-calorie diet.
Generic Orlistat capsules are also used to reduce the risk for weight regain
after prior weight loss.
Generic Orlistat is indicated for obese patients with an initial body mass
index (BMI) greater or equal to 30 kg/m 2 or greater or equal to 27 kg/m
2 in the presence of other risk factors (e.g., hypertension, diabetes,
dyslipidemia).

Recommended Adult Dosage
The dose of Generic Orlistat is one 120 mg capsule three times
a day with each main meal containing fat (during or up to 1 hour
after the meal).
The patient should be on a nutritionally balanced, reduced-calorie
diet that contains approximately 30% of calories from fat. The
daily intake of fat, carbohydrate, and protein should be
distributed over three main meals.
If a meal is occasionally missed or contains no fat, the dose of
Generic Orlistat can be omitted.

Recommended Adult Dosage
Because Generic Orlistat has been shown to reduce the absorption of
some fat-soluble vitamins and beta-carotene, patients should be
counseled to take a multivitamin containing fat-soluble vitamins to
ensure adequate nutrition. The vitamin supplement should be taken at
least 2 hours before or after the administration of Generic Orlistat, such
as at bedtime.
The 60 mg Generic Orlistat capsule is for weight loss in overweight
adults, 18 years and older, when used along with a reduced-calorie and
low-fat diet. Take 1 capsule of 60 mg with each meal containing fat. Do
not take more than 3 capsules daily.

Contraindications

Generic Orlistat is contraindicated in patients with a
known hypersensitivity to Orlistat or any component of
the capsule.

Generic Orlistat is contraindicated in pregnancy

Generic Orlistat is contraindicated in patients with
chronic malabsorption syndrome

Generic Orlistat is contraindicated in patients with
Cholestasis

Storage Instructions
Generic Orlistat Capsules have to be stored at
controlled room temperature i.e. from 20°C to
25°C (68°F to 77°F) with excursions permitted
between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Generic Orlistat Capsules can decrease cyclosporine
exposure.
Take a multivitamin supplement that contains fat-soluble
vitamins to ensure adequate nutrition.
Rare cases of severe liver injury with hepatocellular necrosis
or acute hepatic failure have been reported.
Patients may develop increased levels of urinary oxalate
following treatment with Orlistat capsules. Monitor renal
function in patients at risk for renal insufficiency.

Warnings
Substantial weight loss can increase the risk of
cholelithiasis.
Exclude organic causes of obesity (eg,
hypothyroidism) before prescribing Orlistat Capsules.
Gastrointestinal events may increase when Generic
Orlistat capsules are taken with a diet high in fat
(greater than 30% total daily calories from fat).

Possible Side Effects
Most common Generic Orlistat capsules side effects
with an incidence rate of greater than or equal to 5%
and at least twice that of placebo include oily
spotting, flatus with discharge, fecal urgency
fatty/oily stool, oily evacuation, increased defecation
and fecal incontinence.

Possible Side Effects
Side effects from placebo-controlled clinical trials that occurred at a
frequency of 2% or greater among patients treated with Orlistat
Capsules 120 mg three times a day and with an incidence that was
greater than placebo include Abdominal Pain/Discomfort, Nausea,
Infectious Diarrhea, Rectal Pain/Discomfort, Tooth Disorder, Gingival
Disorder, Vomiting, Influenza, Upper Respiratory Infection, Lower
Respiratory Infection, Ear, Nose & Throat Symptoms, Back Pain, Pain
Lower Extremities, Arthritis, Myalgia, Joint Disorder, Tendonitis,
Headache, Dizziness, Fatigue, Sleep Disorder, Rash, Dry Skin, Menstrual
Irregularity, Vaginitis, Urinary Tract Infection, Psychiatric Anxiety,
Depression, Otitis and Pedal Edema.

Generic Orlistat Capsules Overdosage
Single doses of 800 mg Orlistat Capsules and multiple doses of
up to 400 mg three times a day for 15 days have been studied in
normal weight and obese subjects without significant adverse
findings. Should a significant overdose of Orlistat occur, it is
recommended that the patient be observed for 24 hours.
Based on human and animal studies, systemic effects
attributable to the lipase-inhibiting properties of Orlistat
should be rapidly reversible.

Generic Orlistat Capsules During Pregnancy
Orlistat Capsules has been classified by the US FDA as
Pregnancy Category X.
Orlistat capsules are contraindicated during pregnancy,
because weight loss offers no potential benefit to a pregnant
woman and may result in fetal harm.
A minimum weight gain, and no weight loss, is currently
recommended for all pregnant women, including those who are
already overweight or obese, due to the obligatory weight gain
that occurs in maternal tissues during pregnancy.

Generic Orlistat Capsules During Pregnancy
No embryotoxicity or teratogenicity was seen in animals that
received Orlistat Capsules at doses much higher than the
recommended human dose.
If this medicine is used during pregnancy, or if the
patient becomes pregnant while taking this drug, the
patient should be apprised of the potential hazard of
maternal weight loss to the fetus.
