5. Introduction
The “clot on boiling test”
(COB) is a test done with an
inspection of the warmth
stability of milk through
processing.

6. Principle
The heating of milk in an
advanced state of souring
or abnormal milk will
result in clotting.

7. Precaution
• Avoid direct heating of
milk .
• Use hot water bath

8. Objectives
• To determine the
stability of milk for
heat processing .

Required
• Test tube 20 ml
capacity .
• Hot water bath
• Test tube stand
• Heating arrangement

13. Procedure
1. Take 5 ml of milk in the test tube .
2. Put this on boiling water bath for 5 minutes .

14. 3. Remove the tube from water bath without
shaking .

15. 4. Note any acid smell or precipitated particles
on the sides of the test tube . Sample showing
precipitated particles are recorded as positive
test such milk is rejected on the platform .

Strengths
• Clot on boiling test is a simple and quick
test that can be performed in most
laboratories with little equipment.
Weaknesses
• The test is not specific for coagulation
disorders and may be abnormal in other
conditions.

conditions such as thrombocytopenia.
