Life sciences regulatory authorities in North America, Europe and Japan have been developing standards for drug and biologic registration submissions in electronic format for over ten years. The current version of the format—electronic Common Technical Document (eCTD)—has been the recommended format since 2005 and is on the way to becoming mandatory. This presentation will provide an overview of best practices to enable a sponsor to prepare a compliant eCTD for regulatory authority review. A California biotech submitted an eCTD to the US FDA in summer 2005. This company used commercial-off-the-shelf publishing software, experienced consultants, and in-house staff to compile this eCTD. This drug was the biotechs first candidate for commercialization. The biotech was partnered with big Pharma to assist with sales and marketing after the FDA authorized the drug for marketing. The biotech hired a sales force of nearly 200 in anticipation of FDA approval. The FDA issued a refuse-to-file on this eCTD in fall 2005, due to problems with the PDF files and navigability of the content of the submission. The company’s market capitalization dropped 50% overnight. The company had to recompile the submission, by reworking the source files and rebuilding the XML backbone. The new version of the eCTD was resubmitted in early 2006. The FDA accepted the resubmission for review. However, in May 2006 the FDA issued an approvable letter, that meant the drug could be approved in the future if certain conditions were met (meaning yet another delay in being able to market the product, plus additional expense to conduct further clinical studies). In June, 2006, the big pharma company terminated the partnership with the biotech. In July 2006, the biotech laid off their sales force. In August, 2006, the biotech laid off 100 more employees. In September 2006, the biotech had a meeting with FDA to discuss what additional studies and analysis was needed. Finally, in January 2007, the biotech announced plans to resubmit its NDA by end of second quarter 2007. The U.S. FDA and international regulatory bodies require usability testing of medical devices, products, software, and systems as part of their overall validation. Manufacturers must demonstrate that all potential use-related hazards have been identified, prioritized, and mitigated. The method for demonstrating this is human factors/usability engineering (HF/UE) validation testing. However, the way we conduct these studies is in many ways different from the way we conduct studies of non-medical products and systems. This topic is relevant to the Boston UX community given the convergence of consumer and medical devices, as well as the rise of wearable technologies and the apps that interact with them. This presentation will cover the key aspects of HF/UE validation (a.k.a. ‘summative’) testing and what the FDA expects in the final HF/UE summary report. Importantly, this session will consist of half presentation and half Q&A, with the audience driving the discussion toward current issues, questions, and challenges that are relevant to them.

A device can be any electronic element with some computing ability that supports the installation of firmware or third-party software. DSL Digital Subscriber Line or Digital Subscriber Loop. Refers to the variety of different types of Digital Subscriber Line protocols – high-speed data transmission protocols that are compatible with regular copper telephone wire. DSL is typically used to provide a continuous, high-speed connection directly to an Internet Service Provider Email Electronic mail. A service that sends messages on computers via local or global networks. Home network A local area network (LAN) that connects the PCs in a home and lets users access the Internet simultaneously, share drives, share files and printers, and play head-to-head multi-player games. Internet A network of networks; a group of networks interconnected via routers. The Internet (with a capital I) is the world's largest internet. Internet Access Access to the Internet via a dial-up account via telephone circuit or direct connection. Internet Protocol (IP). The IP part of TCP/IP; the protocol used to route a data packet from its source to its destination over the Internet. IP Address The unique 32 bit number assigned to each computer connected to the Internet and used by the TCP/IP protocol to route packets of data to their destinations. The number is usually written in shorthand "dotted octet" notation in which the 32 bit address is grouped into four sets of 8 bits. Each of those eight-bit sets is converted into a decimal number, and the four resulting decimal numbers are written separated by dots. Most Internet addresses consist of a network portion and a node portion. The address for a host must be unique on the network. When you connect to a web server, for example, you may tell your browser to connect to www.mysite.com, but your computer ultimately has to translate the name to its IP .

See the companion webinar at: http://embt.co/1uHXmjv The ever-growing interest in data accumulation from multiple sources and organizations for reporting and analysis exposes a dirty secret: those business terms that we all think we understand actually have a wide variety of definitions. Sometimes these variances are largely irrelevant, and do not significantly impact the ability to create a reasonable report. However, there are some instances in which even minor variations in structure, content, or semantics can have a significant impact in delivering trustworthy results. This leads to the question: if we have two different structures or definitions for what appear to be two similar concepts, should we harmonize the definitions and structures into one? In some cases this will be a good idea, and it will lead to increased consistency, but this is only true as long as the two concepts really refer to the same real-world idea. In other cases, the same terms are used for two different ideas, necessitating a division into two or more qualified business terms and definitions.
