O documento resume estudos sobre o impacto de plantas geneticamente modificadas (PGMs) resistentes a pragas que produzem proteínas Bt sobre organismos não-alvo, incluindo parasitóides e predadores. Estudos mostraram que a maioria dos parasitóides e predadores testados não foram afetados pela exposição às toxinas Bt em doses similares às encontradas em plantações de milho Bt. Alguns estudos identificaram efeitos sobre poucas espécies quando expostas a altas doses de toxinas Bt.

• Natural do solo, insetos mortos, filoplano,
água, resíduos de grãos etc
• Bactéria Gram positiva, forma esporos e,
produz cristal proteíco durante a fase de
esporulação e ou estacionária
16. Parasitóides
• Tipos de parasitismos:
– Conhecer a espécie que se trabalha –
taxonomia
– Endo e Ectoparasitóides
– Endo – desenvolvem-se dentro do corpo da
lagarta
– Ecto – na parte externa.
17. Parasitóides
• É um organismo normalmente menor do que o
hospedeiro, somente um indivíduo pode matar o
hospedeiro. Exige somente um hospedeiro para
completar o seu ciclo de vida. O adulto tem
vida livre. A maioria dos parasitóides
pertencem a cinco ordem de insetos, mas a
grande maioria dentro das ordens Diptera e
Hymenoptera.

• Observar sempre a qualidade do
hospedeiro – no caso de alimentar o
hospedeiro com toxinas Bt
• Usar o inseto taxonomicamente correto
• Se não houver possibilidade, usar um grupo
taxonomicamente próximo.

23. Predadores
• Organismo de vida livre que durante todo o
ciclo de vida, mata suas presas, normalmente
é maior do que a mesma. Necessita de mais do
que uma presa para completar seu ciclo de
desenvolvimento. Mantideos, aranhas,
percevejos etc. Os predadores variam em
relação ao aparelho bucal: mastigador
(Coleoptera) e picador sugador (Hemiptera).
Sugam as presas e normalmente injetam
toxinas.
28. Bioensaios
• Observar sempre a qualidade da presa
a ser oferecida ao predador
• O presa não pode estar debilitada
• No caso de toxinas Bt, realizar
bioensaios para determinar sub doses
para contaminar, porém não matar os
hospedeiros.

Coccinelidae species:

They feed on artificial diet and need Anagasta eggs to lay fertile
eggs. If it is a Bt toxin, e.g cry1Ab, it will be mixed with artificial
diet, and insects will be allowed to feed for 72h. After this period
insects will be fed on artificial diet + Anagasta eggs.
Comment: You could consider to provide the test diet for a period of
time to every of the ladybird larval stages. Thus you increase
exposure to the test substance. We have done this with Adalia
bipunctata using sucrose solution instead of artificial diet (simply
because there is no diet available for this species) (Alvarez-Alfageme
et al. 2011).
Diet will be weighed and also the amount of the Bt toxin (µg of Bt
toxin/ml).
Comment: I like this idea. It would allow you to really quantify the
amount of diet that was consumed and thus the dose of Cry protein
that was ingested. Make sure that you run a control where diet is
provided without addition of a ladybird. I believe that the diet will
change in weight during the exposure period (it may dry out). You
need to correct for this weight loss.
58. Treatments when Bt toxin is used:
T1- ¼ f Bt toxin (µg of Bt toxin/ml) + diet and Anagasta eggs after 72h
T2 – ½ Bt toxin (µg of Bt toxin/ml) + diet and Anagasta eggs after 72h
T3- 1 Bt toxin (µg of Bt toxin/ml) + diet and Anagasta eggs after 72h
T4- 2X Bt toxin (µg of Bt toxin/ml) + diet and Anagasta eggs after 72h
T5- diet + and Anagasta eggs after 72h
These treatments will be performed with immature and all the life
history will be recorded (mainly the period in each instar). When they
become adults, all reproductive capacity will be evaluated and couples
will be isolated as follow:
Female fed on Bt + male fed on Bt
Female fed on Bt + male (from Control)
Female (Control) + male fed on Bt
Female (Control) + male (Control)
Comments: You could consider to terminate the experiment when the
adults emerge and use adult dry weight as the final measurement
endpoint. Adult weight is an indicator for fecundity.
